As already commented, in standard functionality, automatic determination of storage location in sales order is not possible. Furthermore storage location is not a mandatory filed in creation of sales order.
The business reason for that could be that the Order entry staff does not know where exactly the goods are stored during the creation of a sales order. Furthermore, giving plant etc. at this stage is sufficient to go ahead with the sales process.
As Vamsi has explained, the storage location determination can be configured in dlv item category. In addition you would have to activate field "Storage location determination" in 0VLP if atall you want the determination process to be activated / work.
In Delivery doc type, 0VLK, you can set the rule for determination of St loc. So the components to determine S loc could be different from what Vamsi has advised depending upon the rule set in 0VLK.

I agree with your concept/ logic behind your post.
Say in our plant M1 is stored in 0001, 0002, 0003. In S loc determination, we can only configure that M1 can be picked from one of these 3 locations. E.g. you determine storage loc 0001 as automatic. & actually you would like goods to be picked from 0003, then user has to go to VL02N and change the storage location manually.

I have tested that if in the sales order, storage location is kept blank; and delivery is made with S Loc 0001. Going back to VA02, the storage loc in the sales order is blank; and you can not add / edit this field.
So it could happen that in so Sloc is 0001, in delivery SLoc is 0002. & later you can not change 0001 in to 0002.
